You want the tower to be in order ( lowest numbers on the top getting bigger as you go down) They don't have to be in exact order, they just have to be largest numbers on bottom and small on top. For Ex:

YOu could have 1-3-5-7-11-20-25-34-35-50, 1 being on top and 50 being on bottom. Numbers you place in correct order give you extra points.

You have to sort your tower out before the computer, or else it's an instant lose.

In regards to other players, you get bonus points for completing a level, for putting two consecutive number together, and bonus for the number at the top of the tower (i think its the 400 x the number at top of tower).

So you "win" by getting more points compare to other people.
